| I. Background | |
| CXCR4(C-X-C 趨化因子受體 4 型)是一種在人體內(nèi)廣泛表達(dá)的蛋白質(zhì),屬于 GPCR 家族,具有典型的七次跨膜結(jié)構(gòu)。它的專屬配體是 CXCL12(也稱 SDF-1,基質(zhì)細(xì)胞衍生 因子-1)。在生理狀態(tài)下,CXCR4 在免疫細(xì)胞(如 T 細(xì)胞、B 細(xì)胞)、造血干細(xì)胞以及多種組織中均有表達(dá),主要負(fù)責(zé)調(diào)控細(xì)胞的遷移、歸巢和滯留,在免疫應(yīng)答、胚胎發(fā)育以及造血干細(xì)胞的骨髓錨定中扮演著至關(guān)重要的角色。 當(dāng) CXCL12 與 CXCR4 結(jié)合后,會(huì)引發(fā)受體構(gòu)象改變,進(jìn)而激活細(xì)胞內(nèi)多條信號(hào)通路。首先,受體偶聯(lián)的 G 蛋白被激活,進(jìn)而抑制細(xì)胞凋亡并促進(jìn)存活;同時(shí),PI3K/AKT 和 MAPK/ERK 通路被激活,促進(jìn)細(xì)胞增殖、遷移和相關(guān)基因表達(dá);此外,鈣離子的流動(dòng)也會(huì)被觸發(fā),直接驅(qū)動(dòng)細(xì)胞的趨化運(yùn)動(dòng),使其朝著高濃度 CXCL12 的方向定向遷移。這一完整 的信號(hào)傳導(dǎo)過程被稱為 CXCL12/CXCR4 信號(hào)軸。 |
